Description About Klotho-Derived Peptide 1
Klotho-Derived Peptide 1 (KP1) is a 30-amino-acid peptide derived from the anti-aging protein Klotho; it exhibits significant renoprotective and anti-aging activities and is currently primarily used in scientific research. While it recapitulates some functions of the full-length Klotho protein, it possesses a lower molecular weight, making it easier to synthesize and apply. Treatment with KP1 improved renal function, attenuated structural damage, inhibited tubular cell apoptosis, and preserved mitochondrial integrity in two experimental models. Mechanistically, KP1 enters renal proximal tubular epithelial cells via endocytosis, directly targets the mitochondrial protein ATAD3A (ATPase family AAA domain-containing protein 3A), prevents its degradation, and maintains its function. By interacting with HIGD2A (HIG1 domain family member 2A) and sustaining its mitochondrial expression and function, ATAD3A prevents cytochrome c release and inhibits caspase activation following injury, thereby alleviating tubular cell apoptosis.
